血管性血友病因子(von Willebrand Factor, vWF),是由第12號染色體的短臂所編碼的糖蛋白。vWF可同時與膠原纖維和血小板結合，當血管破裂時大量血小板以vWF為中介，粘附在膠原纖維上，形成血栓，得以止血。在正常的生理條件下,絕大部份由內皮細胞合成,其余則由巨噬細胞生成。前者產生后多儲存在W-P小體（Weibel-Palade body，WPB）內, 后者則儲存在血小板的顆粒中。它與一系列心血管疾病如動脈粥樣硬化、急性冠狀動脈綜合征、心房顫動等均關系密切，vWF是一種重要的血漿成分。vWF在止血過程中主要由兩種作用：①與血小板膜GPⅠb-Ⅸ復合物及內皮下膠原結合，介導血小板在血管損傷部位的黏附；②與因子Ⅷ結合，作為載體具有穩定因子Ⅷ的作用。此外，vWF也能結合GPⅡb-Ⅲa，參與血小板的聚集過程。正常人血漿vWF濃度約為10mg/L。在內皮細胞受刺激或損傷以及集體處于應激狀態時，血漿vWF水平升高。vWF及某些止血與纖溶成分都是血栓性疾病的獨立危險因素。相反，當vWF基因發生缺失、插入點突變、剪切點替換或提前形成轉錄終止信號時，導致血漿vWF量有顯著減低或有質的缺陷，不能完成其正常的止血功能時，即為vWD。
